INFLUENCE OF AZID OF SODIUM, CYANIDE OF POTASSIUM AND FLUORIDE OF SODIUM ON TEST REACTION ON SUPPRESSION OF FOAMING AT SACCHAROMYCES CEREVISIAE

For identification of pollutants, assessment of integrated toxicity biotesting is especially perspective. S. cerevisiae yeast is a convenient model eukaryotic organism for carrying out toxicological researches and an economic alternative to use of animals in biotesting. However the known biotests with use of yeast differ in labor input, demand the special equipment. We have offered express test-reaction with use of S.cerevisiae. It is known that S. cerevisiae as yeast of riding fermentation intensively forms foam on the surface of the fermentable liquid. This ability of S. cerevisiae has been used as a test-response. As a test-object served commercial drug of dry «Saf-Moment» S. cerevisiae yeast («Saf-Neva», Russia). To determine the mechanisms of foaming in the yeast suspension with 2% glucose, including the contribution of fermentation and respiration, inhibitors of these processes were used. We used inhibitors of fermentation and respiration to find out the mechanisms of foaming. The conducted researches have shown that NaF had stronger toxic effect on foam-forming activity of S. cerevisiae, than respiratory poisons: NaN3 and KCN. NaF at concentration of 0.01 g/l caused the expressed toxic action on foaming of S. cerevisiae. At the same time process of foaming developed, but has been oppressed, stretched out more out in time in comparison with control. The maximum volume of foam in suspension was 32.5 % less, than in control. In the presence of 10 g/l of NaF process didn't developed. NaN3 in concentration 0.05 and 0.1 g/l made the stimulating impact on foam-forming activity of S. cerevisiae, at the same time the volume of the formed foam was for 31 and 18.7 % more, than in control. Oppression of foaming was noted at increase in concentration of NaN3 to 1 g/l, but full suppression didn't happen even in suspension to addition of 10 g/l. KCN in concentration of 0.05 and 0.1 g/l didn't exert negative impact on foaming in yeast suspension. At increase in content of KCN to 1 g/l the maximum volume of foam was 14 % lower, than in control, process of foaming came to the end for 20 min. later in comparison with control. KCN had strong toxic effect on foaming in concentration 10 g/l. Intensity was lower, than in control. The maximum volume of foam was 51 % lower, than in control. A more pronounced inhibitory effect on the foaming of sodium fluoride may indicate that the fermentation processes prevail in the test reaction being studied.
