The creation of in vitro model of human BBB, completely identical to the real BBB, was necessary for the possibility of studying the transport and penetration of drugs and xenobiotics and is still very popular and urgent task. In order to reproduce the most close to the real in vitro model of BBB, a microfluidic platform is used, which allows more carefully reproduce the structure of BBB, as well as physiological tangential stress on the surface of endothelial cells (0.3 - 2 PA).
An innovative development was a new class of in vitro models: organs-on-chip, combining the advantages of both in vivo and in vitro models. These so-called chips are micro-fluid devices in which tissues are cultivated in special conditions that best reflect the real microenvironment of these tissues. In this paper, an analytical review of modern scientific and technical, regulatory, methodological literature, affecting the scientific and technical problem investigated in the framework of applied research. It is concluded that the BBB-on-chip models can be used to study complex biological processes, combine the advantages of in vitro and in vivo models, have high predictive power, and be a valuable research tool in addition to the classical in vitro and in vivo methods. |
