The cycle of nitrogen is an essential, integral process in nature. Since amino acids form an important part of life-building and maintenance, nitrogen and its chemical compounds are an integral nutrient and ingredient for all living organisms. Understanding the processes in soil is essential because it helps both to ensure agricultural productivity, to control various, soil-based, unwanted pests and diseases, and, crucially, to limit climate change-related factors directly related to soil. The nitrogen cycle research actuality increase according to climate policy aims for next decades. The one of main reactive nitrogen form is nitrous oxide. The aim of this research is to develop methodology of nitrous oxide isotopes measurements in soil samples under laboratory conditions. The study was divided into three phases. Firstly the theoretical studies of nitrous oxide isotopes research and used measurement methods. Secondly the nitrous oxide isotopes measurements were done in 26 soil samples with aerobic and anaerobic soil moisture regime under laboratory conditions using equipment Piccaro G5131-i. Thirdly the nitrous oxide isotopes data was verified and analyzed and developed methodology for nitrous oxide isotope measurements for soil samples under laboratory conditions. The calculated ??15?????????? and ??15??SP values indicates steady results, with some exceptions, and changes over time. During the development of the study, the information gathered and the results obtained, give improvements in methodology of carrying out measurements of nitrous oxide isotopes under laboratory conditions and set new objectives for further studies. In order to obtain as accurate and reliable data as possible, not only the acquisition and verification of nitrous oxide isotope data is important, but it would also be necessary to perform an accurate calibration of the equipment according to developed methodology.
