REGENERATION POTENTIAL OF TOMATO CALLUS CULTURE
Abstract
The purpose of the research was to study the regenerating potential of tomato callus culture depending on the hormonal composition of the medium, genotype, and age of the explant. Introduction in vitro culture was performed according to the standard method. As a basis, the mediums were served -Murashige and Skoog (MS) and Linsmaier and Skoog (LS). Growth hormones were added in various combinations and concentrations. As initial material, samples of local and foreign selection tomato varieties. As explants, cotyledons, hypocotyl, flower buds, parts of the leaf and stem of a vegetative plant were used. The regeneration efficiency was determined after 5 weeks from the start of cultivation as the ratio of the number of shoots (more than 8 mm in length) to the total number of explants. We have evaluated and optimized conditions in a vitro culture that affected the regeneration of shoots from various organs and tissues of tomato. Studies had shown that morphogenetic potential depended on explant, cultivation conditions, and genotype. The greatest regeneration potential was observed in the Big Star F1 hybrid with the cultivation of 6-daily hypocotyl segments 589.2±14.3%. The optimal nutrient medium for intensive callusogenesis and regeneration for the variety samples BigStar F1, Etude and Zvartnots was MS medium, supplemented with BAP - 3.0 mg/l and IAA - 0.5 mg/l.
