STUDY OF BIOSENSOR RECEPTOR LAYER COMPONENT STABILITY UNDER THE INFLUENCE OF VARIABLE ENVIRONMENT ACIDITY

Abstract

The paper presents the impact of variable environment acidity and low temperature on the stability of biosensor receptor layer component BSA (Bovine Serum Albumin). With the use of UV/Vis spectrophotometer, absorption spectra in range (250-300nm) was taken to study conformational changes of the protein with time after exposition to different factors. The results showed an increase in absorption, associated with the conformational changes of BSA with time. A significant decrease in the absorption intensity was noted after the addition to pure BSA - brain metabolite NAA. Performed studies showed that, the increase in the concentration of the NAA brain metabolite caused a decrease in the pH of aqueous solutions of BSA, which correlates the environmental changes to more acidic ones. For samples with higher acidity, the changes in absorption over time were gentle, which may confirm the increased stability of the biosensor matrix factor in an acidic environment. Comparing the effect of low temperature, it could be seen that temperature 4В°C also influence on the protein stability and showed tendency to maintain receptor layer component stable. The observed trend of change, under the influence of external factors was more evident with time.
